Simple switch from TIRF to widefield illumination An arc lamp coupled to the IX2 microscope is focused on an off-center slit using a wedge prism. Thus the light is focused on the outer edge of the back focal plane of the objective, causing the excitation light to exit the objective beyond the critical angle resulting in Total Internal Reflection. For normal fluorescence observation, the wedge prism and slit are simply removed from the light path via a slider in the illuminator. |

Exchange of excitation wavelength The evanescent field is created using white light from an arc lamp, thus the switch from one excitation wavelength to another is achieved simply by insertion of an excitation filter into the light path. The respective slider has three filter pockets and one empty position. |

High-precision fluorescence turret The high-precision fluorescence turret includes three specially adjusted vacant mirror units that permit dichromatic mirror switching while maintaining the position of the illuminating beam on the back focal plane of the objective. This system makes multi-colour observations easy. The usual adjustment of the lightsource after changing mirror units becomes redundant. Up to six mirror units can be installed. |
